  1. Project background:
    Broccoli, also known as broccoli, is a high-quality health vegetable. In recent years, its production area has grown rapidly in the Yangtze River Delta and coastal provinces and cities in East China. The primary problem faced in broccoli production is excellent hybrid varieties lacking independent intellectual property rights. In the past few years, seeds have been almost occupied by foreign seed companies such as Japan, the United States and Europe. In particular, the price of imported seeds of mid-and late-maturing varieties that can winter in the open field has repeatedly climbed to 20000 yuan/kg, which greatly increased the burden on farmers and seriously hindered the development of broccoli production in my country. Another major problem in broccoli production is that fertilization is unscientific and it is impossible to achieve differentiated investment in fertilizers. The yield, yield and fertilization benefits are low, which affects farmers 'income increase and the farmland environment.
  2. Main contents and results:
    (1) In response to the demand of broccoli varieties for overwintering production in my country, this study successively used self-incompatibility and heterosis techniques to cultivate the medium-maturing "Hulu No. 2"(National appraisal in 2007), extremely late-maturing "Shanghai No. 4"(recognized by Shanghai City in 2005), using male sterile heterosis technology and molecular marker rapid identification of sterile germplasm assisted breeding technology to cultivate late-maturing "Hulu No. 5"(recognized by Shanghai City in 2007) and other high-yield, high-quality mid-and-late-maturing series of hybrid broccoli varieties with independent intellectual property rights. "Hulu No. 2" and "Shanghai No. 4" have participated in the national regional test of new autumn and winter broccoli varieties for three consecutive years, and their average yield and output value rank among the best among the tested varieties. The project acceptance opinions believe that varieties such as "Hulu No. 5" are suitable for domestic and foreign market demand and improve the international competitiveness of products. The entire growth period of the new mid-to-late-maturing varieties in the "Hulu" series is about 80, 110 and 130 days respectively, which can achieve continuous production in autumn and winter in the middle and lower reaches of the Yangtze River. In particular, the yield per mu of Hulu No. 5 exceeds 1500 kilograms, which avoids the phenomenon that existing Japanese varieties are prone to red balls under low wintering temperatures, and the finished product yield is increased by more than 20%.
    (2) The SRAP reaction system for broccoli was established and optimized. The specific amplified fragment M6E7 -700 between the male sterile line and the maintainer line of broccoli was found for the first time at home and abroad. SRAP and corresponding SCAR markers closely linked to the sterile gene of this sterile line were established, laying a foundation for the application of molecular marker-assisted selection of broccoli sterile gene, and differences in nuclear DNA between the Brassica Ogura-CMS sterile line and the maintainer line were discovered for the first time.
    (3) Formulated the first Shanghai City local standard in China,"Technical Regulations for Broccoli Seed Breeding" that utilizes self-incompatibility and hybrid technology, especially using cytoplasmic male sterile lines to carry out hybrid utilization technology of broccoli, which has changed the original defects of low efficiency and low purity of hybrid seeds by artificial bud peeling. A new method to quickly identify the purity of hybrid seeds using SRAP markers has also been developed. The above technical method simplifies the seed production process and ensures that the seed purity reaches 99%. Reduce seed production costs by about 50%.
    (4) Systematically carry out research on the nutrient absorption and distribution laws of broccoli, and establish the effect function of nitrogen, phosphorus and potassium fertilizers on broccoli yield, quality and fertilization profit for the first time in China, forming nitrogen, phosphorus and potassium balanced fertilization and nitrogen fertilizer operations for different production goals of high domestic sales and high export finished product yield., and taking the lead in achieving breakthroughs in key technology of soil testing and formula fertilization based on fertilization models for domestic vegetable crops. The "Automatic Calculation System for Soil Testing and Formula Fertilization of Broccoli" with software copyright has been developed to scientifically guide the fertilization production of new varieties.
  3. Application situation and economic and social benefits:
    This study solved the two bottlenecks of independent intellectual property varieties of broccoli and supporting scientific fertilization, laying a foundation for variety and fertilization for improving the yield and quality of broccoli. A total of 50,000 acres of varieties and supporting good methods have been planted and promoted in Shanghai, Jiangsu, Zhejiang, Anhui, Shandong, Henan, Hubei, Hunan and other provinces and cities, with an additional net benefit of approximately 150 million yuan. It has played a technical supporting role in changing the passive situation of completely relying on imports from Japan and other countries for broccoli production, realizing the independent development of the broccoli industry, promoting farmers 'income and cost reduction, agricultural production and efficiency increase, and meeting the demand of the broccoli market in autumn and winter.
