This project belongs to the field of biology and medicine technology. Based on the properties of several different types of active ingredients/components of traditional Chinese medicine, the project has designed and constructed a variety of new nano-drug delivery systems based on the principles and methods of modern biopharmaceutics, improving the oral bioavailability of poorly soluble traditional Chinese medicine ingredients and promoting the efficacy. More than 10 years of research have been carried out on three aspects: percutaneous absorption of ingredients and targeted delivery of anti-cancer ingredients of traditional Chinese medicine, and innovative results have been achieved:
(1) Developed a variety of new technologies for preparing traditional Chinese medicine nano-drug delivery systems, established a new method for preparing biodegradable polymer PCL-PEO-PCL triblock copolymers in one step reaction; developed a method for modifying lipid nanocarriers with WGA; established a new method for modifying polylactic acid drug-loaded nanoparticles with RGD; established a semi-flow method and its device for measuring the solubility of poorly soluble components in supercritical fluids and applied them to the preparation of solid dispersions of poorly soluble effective components of traditional Chinese medicine; A variety of lipid transdermal nano-drug delivery systems have been constructed; nano-powder technology and nano-carrier technology have been combined to the mineral traditional Chinese medicine realgar and its compound Niuhuangxingxiao pills; the design idea of a multi-component nano-drug delivery system has been proposed for the first time.
(2) A new method for evaluating traditional Chinese medicine nano-delivery systems has been established and a technical system for evaluating traditional Chinese medicine nano-delivery systems has been initially established. For the first time, traditional Chinese medicine fingerprint technology was applied to the evaluation of the preparation process of nano-drug delivery systems; dynamic lipid digestion models were used to establish an in vitro method to evaluate the oral absorption of lipid nanopreparations; an in vivo evaluation method based on microdialysis technology was established; For the first time, fluorescent tracing combined with Nanosight technology was applied to the study of the percutaneous absorption process of nanocarriers.
(3) Clarified the mechanism for improving the biological effects of several active ingredients of traditional Chinese medicine. It improved the oral absorption, percutaneous penetration and injection targeting of more than 10 different types of active ingredients/components of traditional Chinese medicine, such as oridonin, bufaolin, aconitine, frankincense/myrrh volatile oil, and realgar, and clarified the mechanism from in vivo, in vitro, cellular, and overall aspects; found that skin activity has a greater impact on the retention of drugs in the skin.
This project has been authorized to have 7 national invention patents and applied for 1 international patent; a total of 56 papers have been published, including 32 SCI papers with a total impact factor of 111.3. The papers have been cited 311 times at home and abroad, including 193 SCI quotations, and a single article has a maximum SCI citation of 67 times. One paper was selected as a key scientific paper by Global Medical Discovery, an international medical information agency (Key scientific article); trained a total of 24 graduate students; participated in 12 academic conferences and reported 4 international conferences; The research results have been applied by many units such as Jiangsu Province Research Institute of Traditional Chinese Medicine, Anhui University of Traditional Chinese Medicine, Anhui Province Xinxing Drug Development Co., Ltd., Shanghai City Key Laboratory of Functional Materials Chemistry (East China University of Science and Technology), Hubei University of Traditional Chinese Medicine and other units in the research and development of innovative pharmaceutical preparations and achieved good results; Through long-term research and accumulation, young scientific research talents have been cultivated, new discipline growth points and stable research directions have been formed, and the development of traditional Chinese medicine pharmacy has been promoted.
